Mechanism of action

Melanotan II (MT-2) is a synthetic cyclic heptapeptide analog of alpha-melanocyte-stimulating hormone (α-MSH). A lactam bridge between its Asp and Lys residues constrains the core His-D-Phe-Arg-Trp sequence, giving it markedly higher potency than linear α-MSH in melanotropic bioassays. It acts as a non-selective agonist at melanocortin receptors MC1R, MC3R, MC4R and MC5R, and has been used in published preclinical research as a pharmacological tool for studying MC1R signaling in melanocytes and central MC3R/MC4R signaling in rodent models.

Research observations

  • Non-selective agonism at MC1R, MC3R, MC4R and MC5R characterized in receptor studies
  • MC1R-mediated cAMP signaling examined in cultured melanocyte models
  • Central MC4R signaling examined in published rodent research
  • Lactam-bridged cyclic structure confers resistance to proteolysis compared with linear α-MSH
  • Potency of cyclic lactam α-MSH analogs studied in melanotropic bioassays
